Peri.4U® Kit

Peri.4U® are human induced pluripotent stem cell (iPSC)-derived peripheral neurons supporting drug development and discovery. The kit contains 1 vial of cryopreserved, human iPSC-derived Peri.4U® neurons (2 million cells), Neuro.4U Basal Medium A (250 ml) and 2 vials Neuro-Supplement 1 (1.25 ml).

Peripheral neurons play an important role in transmitting signals between the periphery and the central nervous system. Our Peri.4U® represent a new human iPSC-derived neuron standard for drug development. Applications include functional and general neurotoxicity assays (e.g., peripheral neuropathy) as well as for botulinum N toxin tests to be used in cosmetic applications in humans.

Peri.4U® Peripheral neurons are a well-characterized, easy-to-use human iPSC-derived neuron model that is up and running in approximately five days post-thaw and has long term (>3 weeks) viability. Per.4U® is >95% pure, fully-differentiated and post-mitotic neurons. Extensively characterized by immunofluorescence staining and microarray for gene expression analysis, Peri.4U®> shows high expression for neuronal markers (Beta-3-Tubulin, MAP2, NCAM1), synaptic markers (Synaptophysin, Synaptotagmin, Synapsin, Drebrin) and peripheral markers (Peripherin, NTRK1, 2, 3, ISL1, RUNX2 and RUNX1T1). Manual patch clamp analysis shows high sensitivity to TTX with an inward and 2 types of outward currents. Peri.4U®show nice primarily-like action potentials and a subpopulation has spontaneous activity. Peri.4U® have functional glutamate and GABA receptors and respond appropriately to neurotransmitter stimulation.
